Jute (Corchorus olitorius) endophytic fungi Grammothele lineata SDL-CO-2015-1 was found to produce epothilone B, a well-reported anticancer compound. Both the genome and RNA-seq data of G. revealed presence gene clusters. In silico mining using NaPDoS (Natural Product Domain Seeker) identified four genes matching with cluster (Epo D). Epothilone extracted from extracellular extract this fungus XAD-16 resin. Different analytical techniques including thin layer chromatography (TLC), reverse phase high-performance liquid (RP-HPLC), mass spectrometry (MS) were used identify characterize following its extraction. TLC bands visible at 365 nm for both standard B samples identical Rf values. Extracted plate further purified RP-HPLC. Using linear gradient system methanol 0.1% formic acid, LC-MS spectroscopy done RP-HPLC fraction. The monoisotopic (M+H+) = 508.1 Da present in chromatogram. product ions m/z values 320.2 Da, 420.2 typical MS2 fragments B. This finding suggests that fungal endophyte, can be potential source J.
